چکیده

The humoral immune system has the ability to acquire memory. Hence, it can recall a previous encounter with an antigen and consequently mount a efficient, specific, and rapid response to a second challenge by that same antigen. A critical aspect of this acquisition of memory is the establishment of a small pool of memory B cells during the latter phases of the primary immune response. In mice and humans, the key characteristics that distinguish antibodies expressed by memory B cells and naive B cells are that memory B cells express somatically hypermutated antibody V regions, and that memory B cells express antibodies that have higher affinities for antigen as compared to antibodies expressed during the primary immune response. The currently accepted model aimed to explain the recruitment of B cells into the memory compartment suggests that these cells follow a hypermutation and

Ofatumumab Monoclonal Antibody Affinity Maturation Through in silico Modeling

Background: Ofatumumab, an anti-CD20 mAb, was approved in 2009 for the treatment of chronic lymphocytic leukemia. This mAb acts through immune-mediated mechanisms, in particular complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity by natural killer cells as well as antibody-dependent phagocytosis by macrophages. Apoptosis induction is another mechanism of this antibody...

Two waves of memory B-cell generation in the primary immune response.

Memory B cells can be generated independently of germinal center (GC) formation and affinity maturation in Bcl-6-deficient mice, but the contribution of the GC-independent pathway for memory B-cell generation in normal mice remains unknown. To examine this, we administrated anti-inducible co-stimulator (ICOS) mAbs into mice at the onset of GC formation in the primary response. This manipulation...

Background: Chlamydia (C.) trachomatis is an obligate intracellular gram-negative pathogen affecting over 600 million people worldwide with 92 million new cases occurring globally each year. Genital C. trachomatis infection has been recognized as the most common cause of pelvic inflammatory disease leading to severe tubal damage, ectopic pregnancy, hydrosalpinx and infertility. However, the mec...

Development of DOTA-Rituximab kit formulation to be labeled with 90Y for radioimmunotherapy of B-cell Non-Hodgkin Lymphoma

NHL is the most common hematologic cancer in adults. Rituximab is the FDA approved treatment of relapsed or refractory low grade B-cell Non-Hodgkin Lymphoma (NHL). But patients eventually become resistant to rituximab. Since lymphocytes and lymphoma cells are highly radiosensitive, low grade NHL that has relapsed or refractory to standard therapy is treated by RIT in which a beta-emitting radio...
